Methodology:
Real-time Polymerase Chain Reaction (PCR)
Clinical significance:
This assay quantifies T cell receptor excision circles (TREC)present in peripheral blood mononuclear cells (PBMC) or CD3 T cells by real-time PCR as a measure of newly produced naïve T cells (thymopoiesis). It is useful for the evaluation of T cell reconstitution following bone marrow/stem cell transplantation or HAART. For evaluation of transplant patients a pre-conditioning sample should be submitted for comparing post-transplant levels. The method can also be used to evaluate T cell impairment
